It does matter if your computer has been infected. If you start your computer in safe mode you use the minimal amount of services to run your computer and usually when a virus gets into your computer it tries to run when you connect to the internet and it must run as a program.
When booting in safe mode you have no connection to the internet and the program the virus is trying to run will most likely not be opened therefor you save yourself from having more things hacked into.
Simply put only when your computer has a virus you should do this.

Doesn’t really matter, safe mode is used when your computer has problems and such, you don’t have to run it in safe mode.
